Disinfecting leaf with 10% dilution of bleach plus few drops of dish detergent. Note that all bleach is not the same. Household bleach tends to be 6.0-8.5% sodium hypochlorite so be cautious when a protocol says a "10% dilution".

Leaf can be cut into pieces or cultured whole.
Leaf pieces can be placed on the surface of the medium "right side up", "upside down" or "sideways".
There will be swelling of the leaf tissue and formation of small bumps.
